目的测定眼镜蛇毒细胞毒素(CV-CTX)对人癌细胞株的细胞抑制作用,以期能简便准确地反映其抗癌活性。方法应用溴化二苯四偶氮盐(MTT)法测定在CV-CTX作用下体外培养的人胃癌细胞株(SGC-7901)和人肝癌细胞株(SMC)的存活情况和其量效关系。结果(1)SMC、SGC-7901的活细胞数与其分解MTT的量成正线性关系。(2)CV-CTX对SMC、SGC-7901癌细胞株的细胞抑制作用呈良好的量效关系,IC50分别为2.45和1.24μg/ml。结论(1)用MTT法测定癌细胞的增殖和衰减是可靠的。(2)应用MTT法证实CV-CTX对体外培养的人癌细胞的杀伤程度可作为其抗癌活性指标。
Objective To determine the cytotoxicity of cobra venom cytotoxin (CV-CTX) on human cancer cell lines in order to reflect its anticancer activity easily and accurately. Methods The survival of human gastric cancer cell line SGC-7901 and human hepatocellular carcinoma cell line (SMC) cultured under the action of CV-CTX and the dose-response relationship were determined by MTT assay. Results (1) The number of viable cells in SMC and SGC-7901 had a positive linear relationship with the amount of MTT. (2) CV-CTX showed a good dose-response relationship with SMC and SGC-7901 cancer cell lines with IC50 of 2.45 and 1.24 μg / ml, respectively. Conclusion (1) MTT assay of cancer cell proliferation and attenuation is reliable. (2) MTT assay confirmed that CV-CTX can kill human cancer cells cultured in vitro as an indicator of anticancer activity.
