目的:探讨PPARγ基因Pro12Ala多态性与妊娠期糖尿病之间的关系。方法:应用聚合酶链式反应-限制性内切酶技术,对104例妊娠期糖尿病(GDM)妇女和120例正常对照者PPARγ2基因Pro12Ala多态性位点进行基因分型。结果:PPARγ2Pro12Ala多态性的基因型频率PP、PA、AA,在GDM组和正常对照组中分别为92.4%、7.6%、0.0%以及85%、13.3%、1.7%。P等位基因和A等位基因在GDM组和对照组的频率分布分别为96.2%、91.7%和3.8%、8.3%。两组间的基因型和等位基因频率均无显著性差异(P>0.05)。在GDM组中,PPARγ2Pro12Ala多态性与体重指数呈正相关(P<0.01),而与空腹血糖、胰岛素、甘油三脂、总胆固醇无相关性。PPARγ2Pro12Ala多态性可能与GDM妇女肥胖的发生有一定的相关性,但与GDM无明显联系。
Objective: To investigate the relationship between PPARγ gene Pro12Ala polymorphism and gestational diabetes mellitus. Methods: Polymerase chain reaction-restriction endonuclease technique was used to genotype the Pro12Ala polymorphism site of PPARγ2 gene in 104 gestational diabetes mellitus (GDM) women and 120 normal controls. Results: The genotype frequency of PPARγ2Pro12Ala was 92.4%, 7.6%, 0.0% and 85%, 13.3% and 1.7% respectively in the GDM group and the normal control group. The frequencies of P allele and A allele in GDM group and control group were 96.2%, 91.7%, 3.8%, 8.3% respectively. There was no significant difference in genotype and allele frequencies between the two groups (P> 0.05). In GDM group, there was a positive correlation between PPARγ2Pro12Ala polymorphism and body mass index (P <0.01), but not with fasting plasma glucose, insulin, triglyceride and total cholesterol. PPARγ2Pro12Ala polymorphism may be associated with the occurrence of obesity in GDM women, but not with GDM.
