目的 :自行研制抗人雄激素受体 (androgenreceptor,AR)单克隆抗体 (McAb) ,以期达到可替代国外AR多克隆抗体。方法 :采用人雄激素受体合成肽 (hAR -N)交联钥孔戚血蓝素 (KLH)为抗原 ,免疫BALB/C小鼠 ,经用Kohler和Milstein杂交瘤技术成功地获得 4株能稳定分泌抗hAR单克隆抗体 (McAb)杂交瘤细胞株 5 5 ,141,2 0 7,2 5 7。结果 :对 45例前列腺癌组织石蜡切片进行了免疫组化 (IHC)检测 ,同时对自制抗hAR -NMcAb与进口抗AR多抗进行分析比较 ,阴阳总符合率为 91%。结论 :自制抗ARMcAb特异性强 ,灵敏度高 ,结果与Zymed公司产品相同 ,操作简便 ,适于临床应用。
OBJECTIVE: To develop a monoclonal antibody against human androgen receptor (AR) by itself, in order to achieve an alternative polyclonal antibody against AR. Methods: BALB / C mice were immunized with hyaluronan (KLH) as an antigen by human androgen receptor synthesis peptide (hAR-N), and four strains of rhBMP-4 were successfully obtained by using Kohler and Milstein hybridoma Stable secretion of anti-hAR monoclonal antibody (McAb) hybridoma cell lines 5 5,141,2 0 7,2 5 7. Results: Immunohistochemistry (IHC) was used to detect the expression of paraffin sections in 45 cases of prostate cancer. Meanwhile, the positive rate of yin-yang was 91%. Conclusion: The self-made anti-ARMcAb has strong specificity and high sensitivity, and the result is the same as Zymed’s products. It is easy to operate and suitable for clinical application.
