本文采用化学和酶促合成兼用的方法,使用无载体~(32)P-正磷酸(或盐)标记5’-[α-~(32)P]脱氧三磷酸腺苷。产物的比活度达到20—160 Ci/m M,实际收率21—40%,放化纯度94—97%,化学合成中生成的~(32)P-脱氧单磷酸腺苷占总放射性量的69—86.7%。反应所需的二种激酶采用高速离心的方法除去抑制酶反应的硫酸铵。本工作还对影响化学合成的因素进行了一些研究。
In this study, 5 ’- [α- (32) P] deoxyadenosine triphosphate (ATP) was labeled with no carrier of ~ (32) P- orthophosphoric acid (or salt) by a combination of chemical and enzymatic methods. The specific activity of the product reached 20-160 Ci / m M, the actual yield was 21-40% and the radiochemical purity was 94-97%. The total amount of adenosine monophosphate of ~ (32) P generated in chemical synthesis Of 69-86.7%. The two kinases required for the reaction were high-speed centrifugation to remove ammonium sulfate that inhibited the enzyme reaction. This work also made some research on the factors affecting chemical synthesis.
