目的在大鼠处于正畸牙移动后保持阶段,以局部注射淫羊藿的方法,来研究其对牙周组织中骨形成蛋白2(bone morphogenetic protein 2,BMP-2)表达的影响。方法在牙移动模型去除前1 d,实验组分别局部注射浓度为10μg/ml,15μg/ml,20μg/ml的淫羊藿溶液,对照组同时注射同浓度同剂量的生理盐水,实验组和对照组都是每隔2 d注射一次,通过HE和免疫组织化学染色观测分析其对大鼠牙移动后保持阶段牙周组织的改建和BMP-2的表达影响。结果:装置去除后第1 w,对照组的复发距离大于实验组各组复发距离分别为0.130 mm,0.148 mm,0.107 mm。实验组成骨细胞数量比对照组分别增加0.32,0.44,0.21,BMP-2表达平均光密度值比对照组分别增加0.019,0.023,0.014,实验组与对照组比较有显著差异。装置去除后第4周,对照组的复发距离大于实验组复发距离分别为,0.105,0.151,0.077。实验组成骨细胞数量比对照组分别增加0.38,0.63,0.01,BMP-2表达平均光密度值比对照组分别增加0.030,0.045,0.023,实验组和对照组两组比较有显著差异(P<0.05)。结论局部注射中药淫羊藿可以增加大鼠实验性牙移动后保持阶段中牙周组织BMP-2的表达,对骨细胞的生成有促进作用。
OBJECTIVE: To investigate the effect of injecting epimedium on the expression of bone morphogenetic protein 2 (BMP-2) in periodontal tissues of rats after orthodontic tooth movement. Methods One day before tooth movement model was removed, the experimental group were injected with 10g / ml, 15μg / ml and 20μg / ml Epimedium solution respectively. The control group was injected with the same dose of saline, experimental group and control The rats in each group were injected once every 2 days. HE and immunohistochemical staining were used to observe the effect of remodeling and expression of BMP-2 on periodontal tissue during the period of tooth movement in rats. Results: At the first week after device removal, the recurrence distance of the control group was larger than that of the experimental group by 0.130 mm, 0.148 mm and 0.107 mm, respectively. Compared with the control group, the number of osteoblasts increased by 0.32, 0.44 and 0.21, respectively. The average optical density of BMP-2 increased by 0.019, 0.023 and 0.014 respectively compared with the control group. There was significant difference between the experimental group and the control group. At 4 weeks after the device was removed, the recurrence distance of the control group was larger than that of the experimental group (0.105, 0.161 and 0.077 respectively). Compared with control group, the number of osteoblasts in experimental group increased by 0.38,0.63,0.01 respectively, and the average optical density of BMP-2 increased by 0.030,0.045 and 0.023 respectively compared with control group, there was significant difference between experimental group and control group (P <0.05 ). Conclusion Local injection of Epimedium can increase the expression of BMP-2 in periodontal tissues during the period of experimental tooth movement in rats, and promote the formation of osteocytes.
