目的探讨百草枯(paraquat,PQ)对星形胶质细胞合成与释放的非递质物质水平的影响。方法人星形胶质(HA1800)细胞分别暴露于终浓度为0、200、400和800μmol/L的PQ 12、24和48 h,ELISA试剂盒法检测细胞培养上清液中胶质源性神经营养因子(GDNF)和脑源性神经营养因子(BDNF)水平。胆固醇定量检测试剂盒法和Western Blot法分别检测0和400μmol/L的PQ对细胞膜中胆固醇含量和载脂蛋白E(Apo E)表达水平的变化。结果与相应时点的阴性对照组比较,PQ能显著降低细胞膜中胆固醇含量和Apo E表达水平(P<0.05),PQ也能以剂量依赖方式降低细胞培养上清液中GDNF和BDNF水平(P<0.05)。结论 PQ能通过影响星形胶质细胞非递质类物质水平,参与调控突触发生及可塑性,进而参与神经退行性疾病的发生。
Objective To investigate the effect of paraquat (PQ) on the level of non-neurotransmitters in astrocytes synthesis and release. Methods Human astrocyte (HA1800) cells were exposed to PQ 12, 24 and 48 h at final concentrations of 0, 200, 400 and 800 μmol / L, respectively. The glial derived neurons Trophic factor (GDNF) and brain-derived neurotrophic factor (BDNF) levels. Cholesterol quantitative detection kit and Western Blot were used to detect the changes of cholesterol and apolipoprotein E (Apo E) levels in PMCs at 0 and 400 μmol / L, respectively. Results Compared with the negative control group at the corresponding time point, PQ significantly decreased the cholesterol content and the expression of Apo E in the cell membrane (P <0.05). PQ also decreased the levels of GDNF and BDNF in the cell culture supernatant in a dose-dependent manner <0.05). Conclusions PQ can participate in the regulation of synaptogenesis and plasticity by affecting the level of non-neurotransmitter in astrocytes, and then participate in the development of neurodegenerative diseases.
