目的研究细胞因子IL-27在急性肺损伤中发挥的作用和相关机制。方法采用盲肠结扎穿孔术(CLP)建立小鼠急性肺损伤模型。ELISA和荧光实时定量PCR法检测CLP-急性肺损伤小鼠模型的肺组织、支气管灌洗液、外周血单个核细胞及血清中IL-27及其亚单位的表达。同时,小鼠行CLP术前吸入给药IL-27中和抗体,经组织学检测小鼠肺组织炎症水平的改变,并检测支气管灌洗液中IL-6、CXCL10、IFN-γ的水平和小鼠存活率。结果 CLP-小鼠的肺组织和外周单个核细胞中IL-27的2个亚单位(EBI3和p28)均增加,支气管灌洗液和血清中的IL-27水平也较对照组显著增加(P<0.01)。抗IL-27多克隆抗体的使用降低了CLP-急性肺损伤小鼠肺组织的炎症水平,支气管灌洗液中IL-6、CXCL10、IFN-γ水平下降(P<0.05),同时提高了小鼠存活率。结论IL-27在急性肺损伤中发挥重要作用,抑制IL-27的表达可能成为预防和治疗急性肺损伤的新途径。
Objective To study the role and mechanism of cytokine IL-27 in acute lung injury. Methods The model of acute lung injury in mice was established by cecal ligation and puncture (CLP). The expression of IL-27 and its subunits in lung tissue, bronchial lavage fluid, peripheral blood mononuclear cells and serum of CLP-acute lung injury mice model were detected by ELISA and real-time fluorescence quantitative PCR. At the same time, mice were given IL-27 neutralizing antibody by inhalation before CLP, histological changes of lung tissue inflammation were detected and bronchial lavage fluid IL-6, CXCL10, IFN-γ levels and Mouse survival rate. Results The two subunits of IL-27 (EBI3 and p28) in lung tissue and peripheral mononuclear cells of CLP-Mice were increased, and the levels of IL-27 in bronchoalveolar lavage fluid and serum were significantly increased as compared with the control group <0.01). The use of anti-IL-27 polyclonal antibody reduced the level of lung inflammation in BALB / c mice with bronchoalveolar lavage fluid (IL-6, CXCL10 and IFN-γ) in bronchoalveolar lavage fluid Rat survival rate. Conclusion IL-27 plays an important role in acute lung injury. Inhibiting the expression of IL-27 may be a new way to prevent and treat acute lung injury.
