目的探讨复方抗瘤冲剂对S180肉瘤的作用及机制。方法通过体内给药,测定抑瘤率及应用流式细胞仪测定S180肉瘤细胞的凋亡率、细胞内Bax及细胞黏附分子CD54的表达量。结果不同剂量复方抗瘤冲剂均有抑瘤作用,抑瘤率分别为19.76%、26.85%和29.94%,抑瘤率与剂量有正相关性,并且对肿瘤细胞有诱导凋亡的作用。Bax的表达水平用药后有明显升高,细胞周期用药前后没有显著改变。复方抗瘤冲剂对CD54表达也有明显的抑制作用。结论复方抗瘤冲剂的抑瘤活性与其诱导细胞凋亡有关,实验结果表明凋亡是通过上调Bax的表达水平来实现的。
Objective To investigate the effect and mechanism of Fufang Kangliu Granule on S180 sarcoma. Methods The tumor inhibition rate was determined by in vivo administration and the apoptosis rate of S180 sarcoma cells, the expression of Bax and CD54 were determined by flow cytometry. Results Different doses of compound anti-cancer granules had anti-tumor effect, the inhibition rates were 19.76%, 26.85% and 29.94%, respectively. There was a positive correlation between the inhibition rate and dose, and the effect of inducing apoptosis on tumor cells. Bax expression levels significantly increased after treatment, before and after the cell cycle did not change significantly. Compound anti-cancer granules on CD54 expression also had a significant inhibitory effect. Conclusion The anti-tumor activity of Fufang Kangliu granules is related to the induction of apoptosis. The experimental results show that apoptosis is achieved by up-regulating the expression of Bax.
