目的探讨小鼠胚胎早期造血发育阶段Notch1信号的表达。方法采用RT-PCR和Western blot法对小鼠胚胎9.5 d(E9.5)、E10.5、E11.5和E12.5的主动脉-性腺-中肾(Aorta-gonad-mesonephros,AGM)区和胎肝中Notch1信号基因和蛋白的表达水平进行检测。结果 AGM区Notch1基因mRNA的表达量较高,且随孕龄的增大而递减;胎肝中Notch1基因mRNA的表达水平有明显的差异,其中E10.5表达量最低,E11.5表达量最高;在各个时相,AGM区与胎肝中Notch1基因mRNA的表达水平差异均有统计学意义(P<0.05)。Notch1蛋白与Notch1基因mRNA的表达规律相同,但除E9.5外,其他3个时相AGM区与胎肝中Notch1蛋白的表达水平差异均无统计学意义(P>0.05)。结论 Notch1信号在造血祖细胞的发育分化中具有重要作用,规律是由AGM区向胎肝区迁移。
Objective To investigate the expression of Notch1 in early stage of mouse embryo hematopoiesis. Methods Aorta-gonad-mesonephros (AGM) region of mouse embryos at 9.5 days (E9.5), E10.5, E11.5 and E12.5 were detected by RT-PCR and Western blot And fetal liver Notch1 signal gene and protein expression levels were detected. Results The expression of Notch1 mRNA in AGM region was high and decreased with increasing gestational age. The expression level of Notch1 mRNA in fetal liver was significantly different, with the lowest expression of E10.5 and the highest expression of E11.5 The expression of Notch1 mRNA in AGM region and fetal liver was significantly different at all time points (P <0.05). The expression of Notch1 and Notch1 mRNAs were the same, but there were no significant differences in the expressions of Notch1 protein in fetal liver and AGM in other three phases except E9.5 (P> 0.05). Conclusion Notch1 signal plays an important role in the development and differentiation of hematopoietic progenitor cells, and the regular pattern is migration from AGM region to fetal liver region.
