目的探讨成年大鼠长时程轴突缺失对大鼠施万细胞中衰老相关β-半乳糖苷酶(senescenceassociated beta galactosidase,SA-β-gal)表达的影响。方法成年雄性SD大鼠随机分为假手术组、1周失神经组、2周失神经组、4周失神经组、5周失神经组、6周失神经组、7周失神经组和8周失神经组。各组动物进行右侧坐骨神经永久性横断手术或者相应的假手术,根据各组设定的远侧段失神经时间分别获取损伤神经的5mm近侧段和10mm远侧段以及假手术组同侧神经和手术组对侧神经的对应节段,采用SA-β-gal染色检测SA-β-gal的表达,并对8周失神经组大鼠获取的损伤神经标本用SA-β-gal和施万细胞特异性蛋白(S100β)双标染色分析远侧段神经中SA-β-gal表达的细胞定位。结果成年大鼠右侧坐骨神经永久横断后,近侧段神经的远端SA-β-gal表达在2周一过性增加,而远侧段神经中SA-β-gal表达在2周明显增加,并且在更长的失神经过程中保持这种高表达水平。而且,绝大部分SA-β-gal的表达出现在S100β免疫反应阳性的施万细胞中。结论长时程轴突缺失增加成年大鼠施万细胞中SA-β-gal表达。
Objective To investigate the effect of long-term axonal deletion on the expression of senescenceassociated beta galactosidase (SA-β-gal) in rat Schwann cells. Methods Adult male Sprague-Dawley rats were randomly divided into four groups: sham operation group, 1 week denervation group, 2 weeks denervation group, 4 weeks denervation group, 5 weeks denervation group, 6 weeks denervation group, 7 weeks denervation group and 8 Week lost the nerve group. Each group of animals were subjected to permanent transection of the right sciatic nerve or corresponding sham operation. According to the set time of distal denervation, the 5 mm proximal segment and 10 mm distal segment of injured nerve and the ipsilateral nerve of sham operation group SA-β-gal was detected by SA-β-gal staining and SA-β-gal and Schwann Cellular localization of SA-β-gal expression in distal segment nerves using double-stained cell-specific protein (S100β) double staining. Results After permanent right anterior sciatic nerve transection in adult rats, the distal SA-β-gal expression in the proximal segment of the nerve increased transiently at 2 weeks, whereas the expression of SA-β-gal increased significantly in the distal segment at 2 weeks In the course of longer denervation to maintain this high level of expression. Moreover, the vast majority of SA-β-gal expression appeared in S100β immunoreactive Schwann cells. Conclusion Long-term axonal deletion increases SA-β-gal expression in Schwann cells in adult rats.
