目的 观察高三尖杉酯碱 (HHar)诱导急性白血病 (AL)原代细胞凋亡的形态、生化特征 ,并分析量效关系 ,以指导临床治疗。方法 经 HHar作用后的 AL 原代细胞由光镜、电镜观察形态特征 ,DNA凝胶电泳、二苯胺法测定生化特征 ,SSPS软件分析量效关系。结果 HHar作用后 AL原代细胞形态、生化特征均符合典型的细胞凋亡改变 ;其DNA片段化率随 HHar作用时间的延长 ,浓度的增高而增加 ,具有时间——效应及剂量——效应线性趋势。DNA片段化率与坏死率正相关。结论 HHar确可诱导 AL 原代细胞凋亡。其诱导细胞凋亡的能力因剂量、作用时间的不同而不同
Objective To observe the morphological and biochemical characteristics of primary apoptosis induced by homoharringtonine (HHAR) in acute leukemia (AL) and to analyze the dose-response relationship to guide the clinical treatment. Methods Morphological characteristics of AL primary cells after HHar treatment were observed by light microscopy and electron microscopy. The biochemical characteristics were determined by DNA gel electrophoresis and diphenylamine method. The dose-response relationship was analyzed by SSPS software. Results The morphological and biochemical characteristics of AL primary cells were in line with the typical apoptotic changes after HHar treatment. The DNA fragmentation rate increased with the increase of HHar ’s action time and concentration, with the time - effect and dose - response linearity trend. The DNA fragmentation rate is positively correlated with the necrosis rate. Conclusion HHar can indeed induce the apoptosis of AL primary cells.
