我们曾对菊三七[Gynura segetum(Lour.)Merr.]与参三七[Ponax notoginseng(Burk.)F. H. Chen]的止血作用进行过对比研究,证明两者止血效能基本相同,作用原理相似。为进一步探讨其止血作用机制,又研究了两者对豚鼠血小板超微结构的影响。用雄性豚鼠20只,参考并改进EC Chew方法:(1) 取硅化器械自心室抽取回心血,离心分离血小板并制成多血小板血浆(PRP)。按优选法设计高、中、低三个剂量组,加入提取物10%菊三七、参三七注射液,生理盐水作对照,分别使药物与血小板浓度比为15、60、120μg/百万(提取物以含生药量计),置37℃恒温水浴箱内孵育后,离心制成PRP团块。(2) 取PRP团块按电镜制片常规处理,超薄切片,醋酸双氧铀-枸橼酸铝双重染色,电镜观察。
