目的分析肝癌组织中乙型肝炎病毒(HBV)感染患者体内DNA甲基转移酶(DNMT)表达量,探讨肝癌组织中HBV感染与甲基化转移酶表达的关系。方法选取2013年1月间至2014年1月青岛市中心医院放疗科接受治疗的52例肝癌患者,根据肝癌组织中HBV感染情况分为HBV感染组和非HBV感染组,每组26例。采用荧光定量聚合酶链式反应(PCR)方法,检测52例患者肝癌组织内DNMT1、DNMT2、DNMT3a和DNMT3b的mRNA表达量。结果 HBV感染组DNMT1 mRNA表达为1.21±0.24,非HBV感染组为0.83±0.21,HBV感染组DNMT1 mRNA表达高于非HBV感染组,差异有统计学意义(t=6.08,P<0.05);HBV感染组DNMT2 mRNA表达为0.77±0.15,非HBV感染组为0.63±0.12,HBV感染组DNMT2mRNA表达高于非HBV感染组,差异有统计学意义(t=3.72,P<0.05);HBV感染组DNMT3a mRNA表达为1.36±0.25,非HBV感染组为0.94±0.17,HBV感染组DNMT3a mRNA表达高于非HBV感染组,差异有统计学意义(t=7.08,P<0.05);HBV感染组DNMT3b mRNA表达为0.68±0.12,0.49±0.09,HBV感染组DNMT3b mRNA表达高于非HBV感染组,差异有统计学意义(t=6.46,P<0.05)。结论肝癌组织中HBV感染促进甲基化转移酶的表达,从而促进抑癌基因甲基化。
Objective To analyze the expression of DNA methyltransferase (DNMT) in patients with hepatitis B virus (HBV) infection and to investigate the relationship between HBV infection and the expression of methylated transferase in hepatocellular carcinoma. Methods Fifty-two patients with hepatocellular carcinoma admitted to Department of Radiation Oncology, Central Hospital of Qingdao from January 2013 to January 2014 were divided into HBV infection group and non-HBV infection group according to the HBV infection in HCC tissues, with 26 cases in each group. The mRNA expression of DNMT1, DNMT2, DNMT3a and DNMT3b in 52 cases of hepatocellular carcinoma was detected by fluorescence quantitative polymerase chain reaction (PCR). Results The DNMT1 mRNA expression was 1.21 ± 0.24 in HBV infection group and 0.83 ± 0.21 in non-HBV infection group. The DNMT1 mRNA expression in HBV infection group was significantly higher than that in non-HBV infection group (t = 6.08, P <0.05) DNMT2 mRNA expression was 0.77 ± 0.15 in infection group and 0.63 ± 0.12 in non-HBV infection group, and DNMT2 mRNA expression in HBV infection group was higher than that in non-HBV infection group (t = 3.72, P <0.05) The expression of DNMT3a mRNA in HBV infection group was significantly higher than that in non-HBV infection group (t = 7.08, P <0.05). DNMT3b mRNA expression in HBV infection group was significantly higher than that in non-HBV infection group (1.36 ± 0.25 vs 0.94 ± 0.17, (0.68 ± 0.12,0.49 ± 0.09). The DNMT3b mRNA expression in HBV infection group was significantly higher than that in non-HBV infection group (t = 6.46, P <0.05). Conclusion HBV infection promotes the expression of methylated transferase in hepatocellular carcinoma tissue, and promotes the methylation of tumor suppressor gene.
