为了观察IL-1β对新西兰兔下丘脑前列腺素受体EP3亚型表达的影响,本实验用IL-1β复制新西兰兔发热模型,并用免疫组化方法检测兔下丘脑内侧视前区(medial preoptic area,MPO)和正中视前区(median preoptic area,MnPO)前列腺素受体EP3亚型的表达。结果表明:静脉注射IL-1β可引起新西兰兔体温升高,最大体温上升幅度(maximal raise altitude of temperature,△T)及1h体温反应指数(temperatu reresponse index,TRI1)均高于对照组;同时,注射IL-1β后,下MPO和MnPO前列腺素受体EP3亚型的表达明显增强。上述结果提示:MPO和MnPO神经元前列腺素受体EP3亚型表达的增强可能是IL-1β引起发热的中枢机制之一。
In order to observe the effect of IL-1β on the expression of prostaglandin receptor EP3 subtype in hypothalamus of New Zealand rabbits, IL-1β was used to replicate the New Zealand rabbit model of fever and immunohistochemical method was used to detect the hypothalamus medial preoptic area , MPO) and median preoptic area (MnPO) prostaglandin receptor EP3 subtype. The results showed that intravenous injection of IL-1β could cause the body temperature of New Zealand rabbits to rise, the maximal raise altitude of temperature (△ T) and the 1-hour temperature response index (TRI1) were higher than those of the control group. At the same time, After injection of IL-1β, the expression of MPO and MnPO prostaglandin receptor EP3 is significantly increased. These results suggest that the enhanced expression of prostaglandin receptor EP3 in MPO and MnPO neurons may be one of the central mechanisms of IL-1β-induced fever.
