目的:研究汉黄芩素对人肝癌HepG2细胞增殖的影响并探讨其可能的作用机制。方法:以人肝癌细胞株HepG2为研究对象,应用CCK-8法检测汉黄芩素对其增殖的影响;流式细胞术检测细胞凋亡率;Western blot检测细胞中凋亡相关蛋白的表达。结果:与对照组比较,20~320μg/mL的汉黄芩素对HepG2细胞的增殖有显著抑制作用,并呈剂量依赖性。不同浓度的汉黄芩素均可诱导HepG2细胞发生凋亡。Western blot结果表明,汉黄芩素能促进促凋亡蛋白BAX的表达,抑制抑凋亡蛋白BCL-2的表达。结论:汉黄芩素能体外抑制人肝癌HepG2增殖,其作用机制可能与上调BAX蛋白表达水平、下调BCL-2蛋白表达水平诱导细胞发生凋亡有关。
Objective: To study the effect of wogonin on the proliferation of human hepatoma HepG2 cells and to explore its possible mechanism. Methods: Human hepatocellular carcinoma cell line HepG2 was used as the research object. The effect of wogonin on proliferation was detected by CCK-8 assay. The apoptosis rate was detected by flow cytometry. The expression of apoptosis-related protein was detected by Western blot. Results: Compared with the control group, Wogonin 20 ~ 320μg / mL significantly inhibited the proliferation of HepG2 cells in a dose - dependent manner. Different concentrations of wogonin can induce HepG2 cell apoptosis. Western blot results showed that wogonin can promote the expression of pro-apoptotic protein BAX and inhibit the expression of anti-apoptotic protein BCL-2. CONCLUSION: Wogonin can inhibit the proliferation of human hepatocellular carcinoma HepG2 cells in vitro. The mechanism may be related to up-regulation of BAX protein expression and down-regulation of BCL-2 protein expression.
