目的 K-ras基因的突变激活,最常见于胰腺癌、结肠癌和肺癌等肿瘤。国外有人报道人喉鳞状细胞癌的发生与K-ras基因的突变激活没有相关性。本文旨在探讨K-ras基因在喉鳞状癌细胞株Hep-2中的表达及其意义,为喉癌发生与K-ras点突变的相关性研究奠定坚实的基础。方法采用RT-PCR技术检测K-ras基因mRNA在喉癌细胞株Hep-2和胰腺癌细胞株MIAPaCa-2中的表达。结果人胰腺癌细胞株MIAPaCa-2和人喉鳞状癌细胞株Hep-2中K-ras mRNA表达均为强阳性。结论人喉鳞状细胞癌细胞株Hep-2中K-ras基因表达阳性,喉癌的发生可能与其点突变引起的激活有关。
Purpose K-ras gene mutation activation, the most common in pancreatic cancer, colon cancer and lung cancer and other tumors. Foreign reports of human laryngeal squamous cell carcinoma and K-ras gene mutations in the activation has no relevance. This article aims to investigate the expression of K-ras gene in Hep-2 laryngeal squamous cell carcinoma cell line Hep-2 and lay a solid foundation for the study of the correlation between K-ras gene mutation and point mutation in K-ras. Methods The mRNA expression of K-ras gene was detected by RT-PCR in Hep-2 laryngeal carcinoma cell line and in pancreatic cancer cell line MIAPaCa-2. Results K-ras mRNA expression in human pancreatic cancer cell line MIAPaCa-2 and human laryngeal squamous carcinoma cell line Hep-2 were all strongly positive. Conclusions The expression of K-ras gene in human laryngeal squamous cell carcinoma cell line Hep-2 is positive. The occurrence of laryngeal carcinoma may be related to the activation induced by point mutation.
