目的研究S-期相关蛋白2(S-phase kinase associated protein2,Skp2)与肿瘤关系研究进展。方法检索国内外相关文献,综述Skp2的生物学功能及其在细胞周期调控中的作用。结果Skp2不仅能够降解细胞周期调控因子,而且参与细胞周期的调控及细胞增殖。结论Skp2作为泛素蛋白酶体途径的底物识别序列,因能够泛素化降解细胞周期调控抑制因子p27及其它细胞周期调控因子参与细胞周期的调控。在许多肿瘤中都发现有Skp2表达的增高,而且其表达水平与肿瘤的预后、恶性进展都密切相关。
Objective To study the relationship between S-phase kinase associated protein 2 (Skp2) and tumor. Methods The related literatures at home and abroad were retrieved and the biological function of Skp2 and its role in cell cycle regulation were reviewed. Results Skp2 not only degraded cell cycle regulators, but also participated in the regulation of cell cycle and cell proliferation. Conclusion Skp2, as a substrate recognition sequence of ubiquitin proteasome pathway, is involved in the regulation of the cell cycle by ubiquitinating the degradation of cell cycle regulatory inhibitor p27 and other cell cycle regulators. Skp2 expression is found in many tumors, and its expression level is closely related to the prognosis and malignant progression of the tumor.
