目的:优选疱疹康喷雾剂的提取工艺。方法:以盐酸小檗碱含量和出膏率的综合评分为指标,通过正交试验考察提取时间、提取次数、加水量对水提工艺的影响。以芦荟大黄素、大黄酸、大黄素、大黄酚、大黄素甲醚总含量和出膏率的综合评分为指标,利用正交试验考察乙醇体积分数、提取时间、提取次数、乙醇用量对醇提工艺的影响。采用HPLC测定盐酸小檗碱及5个大黄苷元的含量。结果:最佳水提工艺为加12倍量水提取3次,每次75 min;盐酸小檗碱质量分数6.07%。最佳醇提工艺为加10倍量75%乙醇回流提取2次,每次75 min;5个大黄苷元总质量分数1.96%。结论:优选的提取工艺稳定可行,适用于疱疹康喷雾剂的工业化生产。
Objective: To optimize the extraction process of Herpes Kang spray. Methods: Taking the comprehensive score of berberine hydrochloride and the rate of ointment as indexes, the effects of extraction time, extraction times and water addition on the process of water extraction were investigated by orthogonal test. The content of berberine hydrochloride and five rhubarb aglycone were determined by HPLC. Results: The best water extraction process for 12 times the amount of water extracted three times, each 75 min; berberine mass fraction 6.07%. The best alcohol extraction process was adding 10 times the amount of 75% ethanol reflux extraction twice, each 75 min; the total mass fraction of 5 rhubarb aglycone 1.96%. Conclusion: The optimal extraction process is stable and feasible, suitable for the industrial production of Herpes Kang spray.
