目的研究太白银莲花皂甙-1对人脑胶质瘤U251细胞生长的影响及其机制。方法采用四甲基偶氮唑蓝(MTT)比色法检测太白银莲花皂甙-1对U251细胞增殖的影响;应用倒置显微镜和Hochest33342荧光染色、透射电子显微镜观察细胞形态的变化;通过逆转录酶多聚酶链反应(RTPCR)检测与凋亡相关基因Bcl-2 mRNA和Bax mRNA水平的表达变化,Western blot分析Bcl-2和Bax在胶质瘤U251细胞中蛋白的表达情况。结果 U251细胞经太白银莲花皂甙-1诱导后,细胞的增殖明显受到抑制,通过RT-PCR结果证实Bcl-2 mRNA在太白银莲花皂甙-1作用下明显呈低表达而Bax mRNA的表达随时间延长逐渐增高,Western blot检测结果显示太白银莲花皂甙-1作用后,下调抑凋亡蛋白Bcl-2的表达并激活了促凋亡蛋白Bax的表达。结论太白银莲花皂甙-1明显抑制U251细胞的生长活性,且能引起胶质瘤细胞大量凋亡,具有显著的抗肿瘤作用。
Objective To study the effects of Aloe saginata-1 on the growth of human glioma U251 cells and its mechanism. Methods MTT assay was used to detect the proliferation of U251 cells. The morphological changes of U251 cells were observed by inverted microscope and Hochest33342 fluorescent staining. The changes of cell morphology were observed by reverse transcriptase The expression of Bcl-2 mRNA and Bax mRNA were detected by polymerase chain reaction (RTPCR). The protein expression of Bcl-2 and Bax in glioma U251 cells was analyzed by Western blot. Results The proliferation of U251 cells was significantly inhibited by the induction of daidzein, and the expression of Bax mRNA was down-regulated under the action of Taibai lotus saponin-1 by RT-PCR. The results of Western blot showed that the expression of Bcl-2 was down-regulated and the expression of pro-apoptotic protein Bax was activated. Conclusion Polygonatum lotion Saponin-1 significantly inhibits the growth of U251 cells, and can induce apoptosis in glioma cells with significant anti-tumor effect.
