目的检测子宫内膜异位症患者在位内膜中lncRNA尿路上皮癌抗原1(UCA1)的表达情况,探讨其在子宫内膜异位症病因学中的意义。方法选取2015年3-11月在中国医科大学附属盛京医院经病理证实为子宫内膜异位症患者的在位内膜标本30例(其中分泌期13例,增生期17例)为实验组,正常子宫内膜标本20例(分泌期7例,增生期13例)为对照组。采用实时荧光定量聚合酶链式反应(qRT-PCR)技术检测两组子宫内膜组织中UCA1的表达。结果实验组在位内膜、增生期内膜和分泌期内膜中UCA1相对表达量明显高于相同时期对照组,差异均有统计学意义(均P<0.01)。而两组组内增生期与分泌期的子宫内膜中UCA1的相对表达量比较,差异均无统计学意义(均P>0.05)。结论在位内膜中UCA1基因的高表达与子宫内膜异位症密切相关,可能参与子宫内膜异位症的发生发展。UCA1的表达水平可能不受内膜月经周期变化的影响。
Objective To detect the expression of lncRNA urothelial carcinoma antigen 1 (UCA1) in endometriosis of endometriosis patients and to explore its significance in the etiology of endometriosis. Methods 30 cases of endometrial specimens (including 13 cases of secretory phase and 17 cases of proliferative phase) were enrolled in this study from March to November in 2015 in Shengjing Hospital of China Medical University who were pathologically confirmed as endometriosis. , 20 cases of normal endometrium specimens (secretory period in 7 cases, 13 cases of proliferative phase) as the control group. Real-time fluorescent quantitative polymerase chain reaction (qRT-PCR) was used to detect the expression of UCA1 in two groups of endometrial tissues. Results The relative expression of UCA1 in eutopic, proliferative and secretory endometrium in experimental group was significantly higher than that in control group at the same period (all P <0.01). However, there was no significant difference in the relative expression of UCA1 between proliferative and secretory endometrium in both groups (all P> 0.05). Conclusion The high expression of UCA1 in eutopic endometrium is closely related to endometriosis and may be involved in the development of endometriosis. The level of UCA1 expression may not be affected by changes in endometrial menstrual cycle.
