结合电子克隆及RT-PCR获得了甘蔗(Saccharumspp.)花穗中表达的一个SR基因成员。该基因cDNA全长960 bp,包含编码209个氨基酸的开放读码框,gDNA序列长2 966 bp,包含5个内含子。同源性比对表明ScSCL25属于SCL亚家族,命名为ScSCL25(gb:JQ031566)。应用生物信息学工具分析表明:ScSCL25编码蛋白为不稳定亲水性蛋白,含有33个潜在磷酸化位点;无信号肽无跨膜结构域,亚细胞定位于细胞核。该蛋白含有SR基因典型的RRM结构域和SR富集结构域,与玉米等作物的SR蛋白具有较高的同源性。
A SR gene expressed in the spikes of sugarcane (Saccharumspp.) Was obtained by electronic cloning and RT-PCR. The cDNA is 960 bp in length and contains an open reading frame of 209 amino acids. The gDNA sequence is 2 966 bp in length and contains 5 introns. Homology alignment indicates that ScSCL25 belongs to the SCL subfamily and is named ScSCL25 (gb: JQ031566). Bioinformatics tools showed that ScSCL25 encoded protein was an unstable hydrophilic protein and contained 33 potential phosphorylation sites. No signal peptide had no transmembrane domain and subcellular localization was located in the nucleus. The protein contains the SR gene typical RRM domain and SR-rich domain, and corn and other crops SR protein has high homology.
