In Chinese medicine, Chinese aconite has been used as a circulatory stimulant, and its active constituents have been reported to possess cardiotonic action. The present study utilized isolated guinea pig atria preparations to investigate the cardiotonic action of two alkaline hydrophilic contents (Wutou alkalinous hydrophilic content, WHC, and Fuzi alkalinous hydrophilie content, FHC), isolated respectively from the root and danghter root of Aconitum earmichaeli Debx. by column chromatographic technique. Guinea pigs weighing 473±89 g were used. Left and right atria were set up in a 20 ml Krebs-Henseleit solution. The solution was constantly aerated with O_2, at 30±0.1℃, equilibrated for 40 min. The contractons of atria were measured with an isometric transducer connected to a recorder. Right atria were allowedto beat spontaneously and left atria were stimulated with rectangular pulses of 1 ms duration at 1Hz at a voltage approximately 20% above the threshold. Concentration-response curves for WHC and FHC were determined by cumulativc additions of drug to the organ bath.
