目的利用超临界二氧化碳(CO2)萃取法获取柴胡桂枝汤挥发油,并在Fmr1基因敲除小鼠(脆性基因敲除小鼠)模型上观察柴胡桂枝汤挥发油的抗癫痫作用,及其与超氧化物歧化酶(SOD)、丙二醛(MDA)、一氧化氮(NO)的关系。方法将30只30日龄的Fmr1基因敲除小鼠(KO)和30只30日龄的野生型小鼠(WT)分别分成两组(KO空白组和KO用药组,WT空白组和WT用药组),按1.7ml/kg的剂量腹腔注射柴胡桂枝汤挥发油,观察柴胡桂枝汤挥发油对小鼠旷场行为的影响,并取不同部位的小鼠脑组织制成1∶10(重量体积比)的组织匀浆,测定SOD活性及MDA、NO的水平。结果与空白组比较,用药组小鼠在旷场实验中运动的平均速度、总路程、穿过各区的次数减少;脑组织中SOD活性增高,而MDA、NO水平均降低,差异有统计学意义(P<0.05)。结论柴胡桂枝汤挥发油对Fmr1基因敲除小鼠的探索性、兴奋性、运动性均有抑制作用;其抗癫痫作用与清除自由基、阻止过氧化物生成,减少NO的神经毒性相关。
OBJECTIVE To obtain volatile oil of Chaihu Guizhi Decoction by supercritical carbon dioxide (CO2) extraction and to observe the antiepileptic effect of the volatile oil of Chaihu Guizhi Decoction on the Fmr1 knockout mice (Fragile Knockout Mouse) model and its interaction with superoxide (SOD), malondialdehyde (MDA) and nitric oxide (NO). Methods Thirty 30 days old Fmr1 knockout mice (KO) and 30 30 days old wild type mice (WT) were divided into two groups (KO blank group and KO treatment group, WT blank control group and WT treatment group Group). The volatile oil of Chaihu Guizhi Decoction was intraperitoneally injected at a dosage of 1.7ml / kg to observe the effect of volatile oil of Chaihu Guizhi Decoction on the open field behavior in mice. The brain tissues of mice in different parts were made into 1:10 (weight-to-volume ratio) The tissue homogenate was used to measure the activity of SOD and the level of MDA and NO. Results Compared with the blank group, the average speed, the total distance and the number of passing through each area of the mice in the untreated group decreased in the open-field test. The SOD activity in the brain tissue increased, while the levels of MDA and NO in the brain tissue decreased, with statistical significance (P <0.05). Conclusion Chaihu Guizhi decoction volatile oil on Fmr1 knockout mice exploratory, excitability, motility were inhibited; its antiepileptic effect and clearance of free radicals, to prevent the formation of peroxide, reduce the neurotoxicity of NO related.
