目的研究一氧化氮对股骨头微循环的作用及与股骨头血流量的量效关系 ,证实一氧化氮的前体—左旋精氨酸能否成为改善股骨头微循环的有效药物。方法随机采用新西兰兔21只 ,分为三组。右股动脉插管 ,分别将左旋精氨酸(0、30、40、60、80、100mg/kg)、NG -硝基 -左旋精氨酸甲酯(0、5、10、15、20、25mg/kg)、NG -硝基 -左旋精氨酸甲酯(10mg/kg) +左旋精氨酸(0、30、60、80、100mg/kg)注入腹主动脉分叉处 ,然后应用氢廓清技术检测每次用药后的股骨头血流量。结果左旋精氨酸剂量与股骨头血流量存在线性回归关系(F=45.28 ,P<0.01) ,回归方程为YF =11.43 +0.07XF ,相关系数r=0.74。NG -硝基 -左旋精氨酸甲酯剂量与股骨头血流量有线性回归关系(F=30.40 ,P<0.01) ,回归方程为YF =12.64 -0.30XF ,相关系数r= -0.67。应用NG -硝基 -左旋精氨酸甲酯10mg/kg后 ,左旋精氨酸的剂量与股骨头血流量无线性回归关系(F=0.13 ,P=0.72)。结论一氧化氮与股骨头血流量存在剂量依赖关系。一氧化氮的前体—左旋精氨酸能明显增加股骨头微循环的血流量 ,NG -硝基 -左旋精氨酸甲酯可阻断左旋精氨酸的作用。
Objective To study the effect of nitric oxide on the microcirculation of the femoral head and the dose-effect relationship with the femoral head blood flow, and to confirm whether LN-L-arginine could be an effective drug to improve microcirculation of the femoral head. Methods Twenty-one New Zealand rabbits were randomly divided into three groups. Right femoral artery cannulation, respectively L-arginine (0,30,40,60,80,100mg / kg), NG - nitro-L-arginine methyl ester (0,5,10,15,20, 25mg / kg), NG-nitro-L-arginine methyl ester (10mg / kg) + L-arginine (0,30,60,80,100mg / kg) into the bifurcation of abdominal aorta, Profiling technology to detect the femoral head blood flow after each dose. Results There was a linear regression relationship between L-arginine dose and femoral head blood flow (F = 45.28, P <0.01). The regression equation was YF = 11.43 + 0.07XF with a correlation coefficient of 0.74. The regression equation between NG - nitro - L - arginine methyl ester dosage and femoral head blood flow was linear regression (F = 30.40, P <0.01). The regression equation was YF = 12.64 -0.30XF with correlation coefficient r = -0.67. There was no linear regression relationship between dose of L - arginine and femoral head blood flow (F = 0.13, P = 0.72) after administration of 10 mg / kg NG - nitro - L - arginine methyl ester. Conclusion There is a dose-dependent relationship between nitric oxide and femoral head blood flow. Nitric oxide precursor - L-arginine can significantly increase the femoral head microcirculation blood flow, NG - nitro-L-arginine methyl ester can block the role of L-arginine.
