本研究把含有人β干扰素基因的植物表达载体pG21(含有β干扰素基因的信号肽)和pG11通过三亲结合转移到了农杆菌C58C1(pGV2260)中,并与pGV2260发生同源重组而稳定存在于Ti质粒pGV2260上,进一步用此载体把β干扰素基因转移到了烟草细胞中,Southern分析证明了此基因已整合到染色体上,RNA点杂交及NPTⅡ酶分析显示此基因在植物细胞中得到了表达。实验中还发现含有pG21的农杆菌比含有pG11的农杆菌更能有效地转化植物细胞,体外实验表明人β干扰素可以有效提高转化率
In this study, the plant expression vector pG21 (signal peptide containing interferon-β gene) containing human interferon-β gene and pG11 were transferred into Agrobacterium C58C1 (pGV2260) by tri-parental binding and stably existed in homologous recombination with pGV2260 On the Ti plasmid pGV2260, the beta interferon gene was further transferred into tobacco cells using the vector. Southern analysis showed that the gene was integrated into the chromosome. RNA dot blot and NPTII enzyme analysis showed that the gene was expressed in plant cells . Experiments also found that Agrobacterium containing pG21 than Agrobacterium containing pG11 more efficient transformation of plant cells, in vitro experiments have shown that human interferon can effectively improve the conversion rate
