目的:建立同时测定山楂叶中芦丁、金丝桃苷及槲皮素的分析方法。方法:采用多波长RP-HPLC法,色谱柱为Agilent Eclipse XDB-C18柱(4.6mm×150mm,5μm);流动相:乙腈(A)-0.025mol/L磷酸水(B),梯度洗脱依次为0~15min(18%A~18%A),15~30min(18%A~40%A);检测波长为363nm(0~18min),371nm(18~30min);流速:0.9mL/min;柱温:28℃。结果:芦丁、金丝桃苷及槲皮素的线性范围分别为0.3148~1.5740μg、0.4205~2.1024μg、0.0261~0.1305μg。平均加样回收率分别为100.29%、99.42%、98.86%,RSD分别为1.79%、1.46%、1.50%。结论:该方法简便、快速、准确,重现性好,可用于山楂叶中芦丁、金丝桃苷及槲皮素的同时测定。
Objective: To establish a method for simultaneous determination of rutin, hyperoside and quercetin in hawthorn leaves. Methods: Multi-wavelength RP-HPLC method was used. The column was Agilent Eclipse XDB-C18 column (4.6mm × 150mm, 5μm). The mobile phase consisted of acetonitrile (A) -0.025mol / L phosphoric acid The detection wavelength was 363nm (0-18min) and 371nm (18-30min) at flow rate of 0.9mL / min (18% A ~ 18% A) Column temperature: 28 ℃. Results: The linear ranges of rutin, hyperoside and quercetin were 0.3148 ~ 1.5740μg, 0.4205 ~ 2.1024μg and 0.0261 ~ 0.1305μg respectively. The average recoveries were 100.29%, 99.42% and 98.86%, respectively, with RSDs of 1.79%, 1.46% and 1.50%, respectively. Conclusion: The method is simple, rapid, accurate and reproducible. It can be used for the simultaneous determination of rutin, hyperoside and quercetin in hawthorn leaves.
