对广东省惠州市疑似感染马铃薯卷叶病毒(Potato leafroll virus,PLRV)的病株毒源进行分子生物学鉴定,为广东省马铃薯卷叶病的防治提供参考。依据CP基因设计的一对特异引物,以马铃薯总RNA为模板,应用RT-PCR技术,成功扩增出336bp的DNA片段。用BLAST对序列进行同源性分析,用Clustal X构建PLRV的聚类图。结果表明:4个惠州PLRV不同分离物的DNA序列和氨基酸结构具有高度同源性,惠州PLRV分离物与国内外13个PLRV分离物具有高度同源性。广东省惠州市的疑似病株确定为感染PLRV的马铃薯病株。
The molecular identification of the pathogen of Potato leafroll virus (PLRV) suspected to be infected in Huizhou City of Guangdong Province provided a reference for the prevention and cure of potato leaf roll disease in Guangdong Province. According to a pair of specific primers designed by CP gene, total RNA of potato was used as a template and a 336 bp DNA fragment was successfully amplified by RT-PCR. Homology analysis of the sequence was performed by BLAST and Clustal X was used to construct the cluster map of PLRV. The results showed that the DNA sequences and amino acid structures of four PLRV isolates from Huizhou were highly homologous. The PLRV isolates from Huizhou shared high homology with 13 PLRV isolates at home and abroad. Suspected strains in Huizhou City of Guangdong Province were identified as potato diseased plants infected with PLRV.
