目的:探讨人参皂苷Rg1对人脐静脉内皮细胞(HUVECs)基质金属蛋白酶-9(MMP-9)及血管内皮生成因子(VEGF)表达的影响,为临床治疗自身免疫性疾病提供理论及实验依据。方法:以TNF-α10 ng/ml刺激HUVECs,在上述条件中加入不同浓度Rg1(0,0.4,4及40 mmol/L),培养24 h后,分别用实时荧光定量PCR(real-time PCR)及免疫印迹实验(western Blot)检测MMP-9及VEGF的表达。结果:在基因水平,Rg1能抑制由TNF-α诱导HUVECs的MMP-9和VEGF的表达;在蛋白水平,Rg1能显著下调TNF-α诱导HUVECs的MMP-9和VEGF的表达。结论:Rg1通过抑制由TNF-α诱导HUVECs的MMP-9和VEGF的表达,从而在RA疾病治疗过程中发挥作用。
Objective: To investigate the effect of ginsenoside Rg1 on the expression of matrix metalloproteinase-9 (MMP-9) and vascular endothelial growth factor (VEGF) in human umbilical vein endothelial cells (HUVECs), and to provide theoretical and experimental evidences for clinical treatment of autoimmune diseases. Methods: HUVECs were stimulated with TNF-α 10 ng / ml, and different concentrations of Rg1 (0, 0.4, 4 and 40 mmol / L) were added to the above conditions. After cultured for 24 h, And Western Blot to detect the expression of MMP-9 and VEGF. Results: At the gene level, Rg1 could inhibit the expression of MMP-9 and VEGF in HUVECs induced by TNF-α. At the protein level, Rg1 could down-regulate the expression of MMP-9 and VEGF in HUVECs induced by TNF-α. Conclusion: Rg1 plays a role in the treatment of RA by inhibiting the expression of MMP-9 and VEGF in HUVECs induced by TNF-α.
