SNAFL-calcein-AM、rhodamine123分别标记培养的小鼠腹腔巨噬细胞胞浆pH、线粒体pH和线粒体跨膜电位;用激光共聚焦显微镜,观察了巨噬细胞内线粒体的分布,ConA刺激后胞浆pH、线粒体pH及线粒体跨膜电位随时间变化曲线。结果显示ConA刺激可引起线粒体内外pH差增加,线粒体跨膜电位在刺激开始时上升,而后逐渐下降;线粒体质子动力势绝对值在刺激开始一段时间增加,随后逐渐降低到比刺激前更低的水平。
The cytosolic pH, mitochondrial pH and mitochondrial transmembrane potential of cultured mouse peritoneal macrophages were respectively labeled with SNAFL-calcein-AM and rhodamine123. The distribution of mitochondria in macrophages was observed with confocal laser scanning microscope pH, mitochondrial pH and mitochondrial transmembrane potential over time. The results showed that ConA stimulation caused an increase in pH difference between mitochondria inside and outside, and mitochondrial transmembrane potential increased at the beginning of stimulation and then gradually decreased. The absolute value of mitochondrial proton motility increased at a certain period of stimulation and then gradually decreased to a lower level than that before stimulation .
