目的探讨胰高血糖素对胰岛β细胞系——MIN6细胞胰岛素分泌的调控作用。方法体外培养胰岛β细胞系——MIN6细胞,在无糖、低糖和高糖环境下分别给予不同浓度胰高血糖素干预处理,采用酶联免疫吸附剂测定(ELISA)法检测MIN6细胞胰岛素释放量,所得数据进行统计分析。结果无糖环境:1000、500、100ng/L胰高血糖素组的胰岛素量均高于空白对照组,1000ng/L组高于500和100ng/L组;低糖(2.8mmol/L)环境:1000、500ng/L组胰岛素量均高于空白对照组,1000ng/L组高于100ng/L组;高糖(16.7mmol/L)环境:1000、500ng/L组胰岛素量均高于空白对照组,1000ng/L组依次高于500、100ng/L组,以上差异均有统计学意义,P<0.05;在无糖、低糖和高糖环境下,胰高血糖素干预浓度与胰岛素释放量均具有正相关关系(相关系数分别为:无糖:r=0.935,低糖:r=0.967高糖:r=0.999,P均<0.01)。结论胰高血糖素在无糖、低糖和高糖任一环境中均以浓度梯度的形式促进β细胞胰岛素的分泌。
