目的探讨2型糖尿病(T2DM)患者及大血管病变患者外周血CD4+CD28-T细胞及其肿瘤坏死因子(TNF)-α、干扰素(IFN)-γ的变化及其意义。方法应用免疫荧光、流式细胞术及ELISA法检测36例初诊单纯T2DM(A组)、30例T2DM合并大血管病变(B组)和28例正常人(C组)的外周血CD4+CD28-T细胞亚群及血清IFN-γ、TNF-α的水平。结果B组CD4+CD28-T细胞和血清TNF-α、IFN-γ水平均高于A组(P<0.05),且两组均高于C组(P<0.05)。结论自身反应性CD4+CD28-T细胞可通过高分泌IFN-γ等细胞因子参与T2DM及大血管病变发生和发展。
Objective To investigate the changes and significance of CD4 + CD28-T cells, tumor necrosis factor (TNF) -α and interferon-γ in peripheral blood of patients with type 2 diabetes mellitus (T2DM) and macroangiopathy. Methods Immunofluorescence, flow cytometry and ELISA were used to detect the percentage of CD4 + CD28 + T cells in 36 newly diagnosed T2DM patients (group A), 30 T2DM patients with macrovascular disease (group B) and 28 healthy controls (group C) T lymphocyte subsets and serum IFN-γ, TNF-α levels. Results The levels of CD4 + CD28-T cells and serum TNF-α and IFN-γ in group B were significantly higher than those in group A (P <0.05), and both were higher than those in group C (P <0.05). Conclusion Autoreactive CD4 + CD28-T cells may participate in the development of T2DM and macroangiopathy through the secretion of IFN-γ and other cytokines.
