目的筛选适合纯化东北铁线莲总黄酮的大孔吸附树脂工艺。方法采用静态吸附/解吸与动态吸附/解吸相结合的方法,以解吸率为主要指标考察各因素对东北铁线莲总黄酮大孔吸附树脂纯化工艺的影响。结果 6种树脂中D-10l型大孔吸附树脂最适合纯化东北铁线莲总黄酮,其最佳工艺为:上样药液东北铁线莲总黄酮浓度相当于原生药0.2 g/ml,吸附速率为2 BV/h,解吸液乙醇浓度为95%,解吸速率为2 BV/h,最佳上柱药液pH值为4,洗脱剂用量为8倍柱体积。经大孔吸附树脂分离纯化后,东北铁线莲总黄酮的纯度由8.39%提高到24.66%。结论 D-10l型大孔吸附树脂对东北铁线莲总黄酮具有较好的纯化效果。
Objective To screen a macroporous resin suitable for the purification of total flavonoids from Clematis verbena. Methods The combination of static adsorption / desorption and dynamic adsorption / desorption was used to investigate the effect of various factors on the purification process of the macroporous adsorption resin of Clematis verbena Thunb. Results D-10l macroporous resin was the most suitable for the purification of the total flavonoids of Clematis natans from 6 kinds of resins. The optimum conditions were as follows: the total flavonoids concentration of the clematis in the samples was 0.2 g / ml, The rate of 2 BV / h, the desorption ethanol concentration of 95%, desorption rate of 2 BV / h, the best on the column liquid pH value of 4, the eluent dosage of 8 column volumes. After the macroporous resin separation and purification, the purity of the total flavonoids of Clematis northeast increased from 8.39% to 24.66%. Conclusion D-10l macroporous resin has good purification effect on the total flavonoids of Clematis verbenii.
