目的:研究金雀异黄酮对体外培养股骨组织(骨干和骨骺端)吸收活性和骨代谢的影响。方法:体外分离培养大鼠股骨组织分别培养骨干和骨骺端48 h后采用终浓度为1×10-5 mol·L-1的金雀异黄酮对体外培养骨干和骨骺端进行处理;测定抗酒石酸酸性磷酸酶(tartrate-resistant acid phosphatase,StrACP)、培养基中葡萄糖(glucose,Glu)、乳酸(lactic acid,La)、Real-Time RT-PCR抗酒石酸酸性磷酸酶(tartrate-resistant acid phosphatase,StrACP),集落刺激因子(macrophage colony stimulating factor,MCSF),组织激酶K(cathepsin K,CTSK)。结果:1×10-5 mol·L-1金雀异黄酮可抑制StrACP活性,增加培养基中La含量和减少培养基中葡萄糖含量,抑制StrACP,MCSF,CTSK mRNA的表达。结论:金雀异黄酮抑制体外培养大鼠股骨组织的骨骺端和骨干吸收活性和促进其代谢活性。
Objective: To study the effects of genistein on the absorption activity and bone metabolism of cultured femoral bone tissue (the trunk and metaphysis). METHODS: In vitro cultured rat femur tissues were cultured for 48 h and then cultured in vitro with genistein at a final concentration of 1 × 10-5 mol·L-1, respectively. The anti-tartaric acid Tartrate-resistant acid phosphatase (StrACP), glucose, Glu, lactic acid (La), Real-Time RT-PCR tartrate-resistant acid phosphatase ), Macrophage colony stimulating factor (MCSF) and cathepsin K (CTSK). Results: 1 × 10-5 mol·L-1 genistein could inhibit the activity of StrACP, increase the content of La in the medium and decrease the glucose content in the medium, and inhibit the expression of StrACP, MCSF and CTSK mRNA. CONCLUSION: Genistein inhibits the absorption of metaphyseal and diaphyseal ends of femur and the metabolic activity of osteoclasts in vitro.
