目的 研究四环素 -哌嗪雌酚酮 (XW6 30 )对胚胎小鼠骺板 型胶原的表达的影响。方法 采用胚胎小鼠体外培养模型 ,通过免疫组织化学手段 ,观察 XW6 30对 型胶原蛋白表达水平的影响。结果 当培养基中XW6 30浓度为 10 - 7m ol/L和 10 - 8mol/L时 ,各区 型胶原蛋白免疫组织化学染色阳性细胞面积与对照组相比均显著增加 ;与同浓度的雌酚酮组相比 ,10 - 7mol/L XW6 30组增殖区和肥大区阳性细胞面积增加。当浓度降至 10 - 9mol/L时 ,只有肥大区阳性细胞面积增加。随浓度升高 ,各区阳性细胞面积呈增加趋势。结论 在体外培养条件下 ,XW6 30剂量依赖地上调小鼠骺板静止区、增殖区和肥大区 型胶原蛋白的表达水平。
Objective To investigate the effect of tetracycline-piperazin-17-ol (XW6 30) on the expression of epiphyseal plate collagen in embryonic mice. Methods Embryonic mouse in vitro culture model was used to observe the effect of XW6 30 on collagen type Ⅰ expression by immunohistochemistry. Results When the concentration of XW6 30 in culture medium was 10-7 mol / L and 10 - 8 mol / L, the area of positive cells in each type of collagen immunohistochemical staining increased significantly compared with that of the control group. With the same concentration of estrone Compared with the control group, the area of positive cells in 10 - 7mol / L XW630 group was increased. When the concentration dropped to 10 - 9mol / L, only the area of positive cells increased. With the increase of concentration, the area of positive cells showed an increasing trend. Conclusion In vitro culture, XW630 dose-dependently up-regulated the expression of collagen in the static zone, proliferative zone and hypertrophic zone of the epiphyseal plate in mice.
