目的采用HPLC法测定大青叶中的靛蓝。方法采用DiamonsilTMC18柱(150 mm×4.6 mm,5μm),检测波长280nm,柱温40℃,考察不同色谱条件下靛蓝的色谱行为;利用正交设计研究靛蓝的超声提取工艺,用优化的HPLC条件测定靛蓝的含量。结果靛蓝1.33~42.56μg.mL-1与峰面积的线性关系良好(r=0.9995),平均回收率为97.7%(RSD=1.7%,n=9)。结论所建方法简便、快速,适用于大青叶的质量控制。
Objective To determine indigo in Folium Isatidis by HPLC. Methods Diamonsil TMC18 column (150 mm × 4.6 mm, 5 μm) was used. The detection wavelength was 280 nm and the column temperature was 40 ℃. The chromatographic behavior of indigo under different chromatographic conditions was investigated. The orthogonal design was used to study the ultrasonic extraction of indigo. Indigo content. Results The linear range of 1.33 ~ 42.56μg.mL-1 was linear with the peak area (r = 0.9995). The average recovery was 97.7% (RSD = 1.7%, n = 9). Conclusion The proposed method is simple and rapid and suitable for the quality control of Folium Isatidis.
