目的:探索用声光可调(AOTF)-近红外光谱(NIRS)技术在线测定丹参提取过程中丹参酮ⅡA含量的方法。方法:收集丹参提取过程中的样品溶液,以HPLC测定其中丹参酮ⅡA的含量,运用偏最小二乘法(PLS1)建立NIRS预测值与HPLC分析值之间的校正模型,对未知样品进行含量预测来验证所建模型。结果:建立的定量模型准确性较好,丹参酮ⅡA的内部交叉验证均方差为RMSECV=0.0092,决定系数是R2=0.9918,校正模型对验证集样品的验证结果为相对误差5.74%。结论:AOTF-NIRS法测定丹参提取过程中丹参酮ⅡA含量,方法快速、无损、预测结果较好,为中药提取过程分析及在线质量控制奠定了基础。
Objective: To explore a method for online determination of tanshinone Ⅱ A during the extraction of Salvia miltiorrhiza with acousto - optic tunable (AOTF) - near infrared spectroscopy (NIRS). Methods: The sample solution was collected during the extraction of Salvia miltiorrhiza. The content of tanshinone Ⅱ A was determined by HPLC. The calibration model between NIRS predictive value and HPLC analytical value was established by partial least squares (PLS1), and the content of unknown sample was verified Model built. Results: The quantitative model was established with good accuracy. The mean square error of cross-validation of tanshinone Ⅱ A was RMSECV = 0.0092 and the coefficient of determination was R2 = 0.9918. The validation result of the calibration model for the validation set was 5.74%. Conclusion: AOTF-NIRS method for the determination of tanshinone Ⅱ A content during the extraction process of Salvia miltiorrhiza, the method is rapid and nondestructive, the prediction result is good, which laid the foundation for the extraction process analysis and online quality control of the traditional Chinese medicine.
