本研究以我国160份来源于温带、热带与亚热带玉米自交系为材料,根据已经公布的D8基因序列及已鉴定的677功能位点,开发CAPS标记。结果发现限制性内切酶Bss SⅠ可用于该位点酶切分型,15个样品的分型结果与测序结果完全一致;利用开发的CAPS标记,对160份玉米自交系D8基因677位点进行基因分型,结果表明在160份自交系中共有151份能扩增出清晰条带,在9份自交系未扩增到条带;20份自交系D8基因677位点基因型为G型,131份自交系基因型为C型。该结果与Thornsberry等(2001)研究结果中该位点基因型的趋势一致,D8基因677位点开发的CAPS标记可用限制性内切酶快速检测,在玉米分子育种中有潜在的利用价值。
In this study, 160 inbred lines derived from temperate, tropical and subtropical maize were used as materials to develop CAPS markers based on published D8 gene sequences and identified 677 functional sites. The results showed that restriction endonuclease Bss SⅠ could be used for restriction enzyme digestion at this locus. The typing results of 15 samples were completely consistent with the sequencing results. Using the developed CAPS markers, 677 inbred lines of D8 gene The results showed that 151 out of 160 inbred lines could amplify the clear bands and did not amplify in 9 inbred lines. The inbred line D8 677 locus genotype For G type, 131 inbred lines genotype C. This result is consistent with the trend of the genotype at this locus in Thornsberry et al. (2001). The CAPS marker developed at locus 677 of D8 gene can be rapidly detected by restriction endonucleases and has potential value in maize molecular breeding.
