目的:制备一种可以通过检测尿液中HIV抗体来进行无创伤HIV感染的胶体金免疫层析快速诊断试纸条。方法:利用柠檬酸三钠还原法制备直径38 nm的胶体金溶液,利用HIV-1 gp41抗原标记胶体金,制备胶体金免疫复合物,组装胶体金免疫层析快速诊断试纸条。结果:胶体金颗粒粒径均一,胶体金免疫复合物性能稳定,特异性强,试纸条检测时间只需10~20 min。结论:胶体金免疫层析试纸条使用简便,检测快速,结果准确,无需特殊的仪器设备,性价比高,适用于基层医疗单位使用和对大量人群的规模筛查等。
OBJECTIVE: To prepare a rapid test strip of colloidal gold immunochromatography for the detection of at-risk HIV infection by detecting HIV antibodies in urine. Methods: The colloidal gold solution with a diameter of 38 nm was prepared by the method of trisodium citrate reduction. The colloidal gold was labeled with HIV-1 gp41 antigen to prepare colloidal gold immune complex, and the colloidal gold immunochromatographic test strip was assembled. Results: The colloidal gold particles were uniform in size and the colloidal gold immune complexes were stable and specific. The test strip test time was only 10-20 min. Conclusion: The colloidal gold immunochromatographic test strips are easy to use, rapid and accurate, without special equipment, cost-effective, suitable for the use of primary medical units and large-scale screening of large populations.
