目的:本研究旨在分析孕鼠暴露于全氟辛烷磺酸(potassium perfluorooctanesulfonate,PFOS)后对仔鼠脑组织蛋白成分的影响。方法:将孕3 d母鼠随机分为2组。正常组孕鼠给予溶剂生理盐水灌胃;染毒组孕鼠给予0.5 mg/kg PFOS灌胃染毒,染毒连续进行7 d。收集正常组所产仔鼠(A组)与0.5 mg/kg PFOS染毒组所产仔鼠(B组)出生后24 h内全脑组织,使用蛋白组学shotgun法进行脑组织蛋白成分鉴定并分析。结果:鉴定结果显示染毒组所产仔鼠脑组织蛋白种类相比于正常组数量减少,且有无功能蛋白产生。结论:孕鼠PFOS染毒对其仔鼠脑内蛋白质的种类组成有明显影响。
Aims: This study aimed to analyze the effects of potassium perfluorooctanesulfonate (PFOS) on the protein composition in the brains of pregnant rats. Methods: Pregnant 3 d female rats were randomly divided into two groups. Rats in normal group were given solvent saline for intragastric administration. Pregnant mice in the exposure group were given intragastric administration of 0.5 mg / kg PFOS for 7 days. Whole brain tissues were collected within 24 h after birth in the offspring of normal mice (group A) and offspring of 0.5 mg / kg PFOS exposed groups (group B), and proteomic analysis was performed using the shotgun proteomic method analysis. Results: The identification results showed that the number of protein in the brain tissue of offspring rats in the exposed group decreased compared with the normal group, and there was no functional protein. Conclusion: PFOS exposure in pregnant rats has a significant effect on the protein composition in the brain of offspring rats.
