目的 构建链置换扩增 (SDA)压电DNA传感器 ,研究其响应特性。方法 ①将SDA与压电DNA传感器结合 ,建立SDA压电DNA传感器结合技术 ;②以结核菌IS61 1 0片段为检测对象 ,研究SDA压电DNA传感器的响应特性。结果 ①SDA压电DNA传感器的响应信号达到 50 0Hz左右 ;②SDA压电DNA传感器出现频率下降的时间长度 (响应时间 )与加入的检测核酸的浓度相关 ;③不同浓度检测核酸 (靶核酸 )导致的频率下降绝对值差别不大。结论 SDA压电DNA传感器能直接检测基因组DNA ;其响应时间与检测物 (靶核酸 )的加入量相关 ,可以用作定量依据
Objective To construct a strand displacement amplification (SDA) piezoelectric DNA sensor and study its response characteristics. Method 1 SDA was combined with piezoelectric DNA sensor to establish the SDA piezoelectric DNA sensor binding technology. 2 The IS6110 fragment of M. tuberculosis was used as the test object to study the response characteristics of SDA piezoelectric DNA sensor. Results 1 The response signal of the SDA piezoelectric DNA sensor reached about 50 Hz; the length of time (response time) of the frequency of appearance of the 2SDA piezoelectric DNA sensor was related to the concentration of the added detection nucleic acid; 3 the frequency of detection of nucleic acid (target nucleic acid) at different concentrations The difference in the absolute value of the decline is not significant. Conclusion The SDA piezoelectric DNA sensor can directly detect genomic DNA; its response time is related to the amount of the analyte (target nucleic acid) and can be used as a quantitative basis.
