以12 5Ⅰ -心得静为放射配基 ,采用冰冻切片技术建立了对大鼠心肌 β肾上腺素能受体 (adrenoceptor,AR)的测定法。实验结果证明 ,心肌AR与放射配基的特异结合在12 5Ⅰ -心得静浓度为 40 0pmol/L时达饱和 ,并确定在 12 5~ 80 0pmol/L范围为最适配基浓度。本法消耗组织微量 ,并可在近似生理条件下研究受体的特征变化。
The 125 I - I was used as the radioligand and the frozen section was used to establish the rat cardiac adrenoceptor (AR) assay. The experimental results show that the specific binding of cardiac AR to radioligand saturates at a concentration of 40 pmol / L at 12 5 I-level and the optimum concentration is 125 to 80 pmol / L. This method consumes tissue micro-volumes and studies the changes in receptor characteristics under approximate physiological conditions.
