研究全反式维甲酸 (RA)在体外诱导人髓系白血病细胞系 (HL 60 )分化过程中细胞周期的改变。利用流式细胞术动态测定RA在体外诱导HL 60细胞分化过程中细胞周期的改变。结果发现 :①在RA与HL 60细胞共培养 4 8小时内 ,S +G2 /M期比例无明显变化 ,但S期细胞的比例改变与RA的浓度有关 ,在 10 - 6 mol/LRA浓度S期细胞比例先出现一过性升高 ,继而持续下降 ,在 10 - 5mol/L浓度S期细胞比例表现为持续性下降 ;②重培养试验证明 :S +G2 /M和S期细胞比例的下降与成熟分化细胞的增加相一致 ;③RA诱导HL 60细胞成熟分化并不是在一个时间点上同时进入分化状态 ,而一旦细胞开始进入成熟分化 ,即使在无RA存在的情况下 ,细胞仍能分化至成熟。结论提示 :RA与HL 60细胞必须相互作用一定时间后才能触发细胞成熟分化 ,S期细胞的比例改变与RA药物浓度有关 ,一旦细胞开始进入成熟分化 ,即使在无RA存在的情况下 ,细胞仍能分化至成熟
