目的观察镉对卵泡生长发育和性激素分泌影响,探讨镉对未成年雌性大鼠卵巢发育的毒性作用。方法未成年雌性清洁级Wistar大鼠48只,按体重随机分为4组,对照组(生理盐水)和低、中、高剂量镉染毒组(0.1、0.3、0.9 mg/kg),腹腔注射染毒,1次/d,连续19 d;染毒结束后,捡测大鼠血清雌激素(E2)、孕激素(P4)水平;取出大鼠卵巢组织连续切片观察卵巢各级卵泡数目构成比。结果低、中、高剂量染镉组血清中E2的水平分别为(6.04±0.28)、(6.10±0.13)和(5.96±0.18)pg/mL,均低于对照组的(6.62±0.24)pg/mL,差异均有统计学意义(P<0.05);高剂量染镉组大鼠各期卵泡数目构成比与对照组比较,差异均有统计学意义(P<0.05)。结论在本实验条件下低剂量镉暴露对未成年雌性大鼠的卵巢发育具有毒性作用。
Objective To observe the effects of cadmium on follicular growth and sexual hormone secretion and explore the toxic effects of cadmium on ovarian development in immature female rats. Methods Forty-eight female Wistar rats were divided into 4 groups randomly according to body weight: control group (saline) and low, medium and high dose cadmium exposure groups (0.1, 0.3, 0.9 mg / kg) The rats were sacrificed and the rats were sacrificed once a day for 19 days. After the treatment, the levels of serum estrogen (E2) and progesterone (P4) were determined. The number of ovarian follicles . Results The serum levels of E2 in low, middle and high dose cadmium groups were (6.04 ± 0.28), (6.10 ± 0.13) and (5.96 ± 0.18) pg / mL, respectively, which were lower than those of the control group (6.62 ± 0.24) pg / mL, the differences were statistically significant (P <0.05). The number of follicles in high-dose cadmium group was significantly higher than that in control group (P <0.05). Conclusions Exposure to low doses of cadmium under this experimental condition has toxic effects on ovarian development in juvenile female rats.
