用正常人淋巴细胞,总RNA与人垂体生长激素特异性探针进行Northern杂交,得到一条弱的杂交带,其位置与人垂体生长激素分泌瘤总RNA的Northern杂交带近乎一致。根据人垂体生长激素cDNA顺序设计合成寡核苷酸引物,用逆转录。聚合酶链反应相结合的方法,从正常人外周血淋巴细胞.总RNA中扩增出的cD-NA与人垂体生长激素cDNA大小相似,证实正常人淋巴细胞有免疫反应性生长激素mRNA的表达。
Northern blotting with normal human lymphocytes, total RNA and human pituitary growth hormone-specific probe resulted in a weak hybridization band whose location was approximately the same as that of the total RNA of human pituitary growth hormone secreting tumors. According to the sequence of human pituitary growth hormone cDNA, oligonucleotide primers were synthesized and reverse transcribed. Polymerase chain reaction is a combination of methods from normal human peripheral blood lymphocytes. The cD-NA amplified from total RNA is similar in size to the human pituitary growth hormone cDNA, confirming the expression of immunoreactive growth hormone mRNA in normal human lymphocytes.
