目的:探讨安定对学龄前儿童滴注氯胺酮的药代动力学影响。方法:16例3.5~6岁择期手术学龄前儿童,随机分为两组,Ⅰ组为氯胺酮组,Ⅱ组为氯胺酮加安定组。用反向高效液相色谱法测定氯胺酮血药浓度,并计算药代动力学参数。结果:两组房室模型均拟合为二房室开放模型、药代动力学参数:Ⅰ组T1/2α=(0.15±0.04)h,T1/2β=(2.84±0.44)h,CL=(0.34±0.06)L/kg·h,Vd=(0.66±0.29)L/kg。Ⅱ组T1/2α=(0.14±0.06)hT1/2β=(3.92±0.52)h,CL=(0.26±0.05)L/kg·h,Vd=(0.65±0.07)L/kg。结论:安定使氯胺酮清除半衰期延长,清除率降低
Objective: To investigate the effects of diazepam on pharmacokinetics of ketamine in preschool children. Methods: Totally 16 preschoolers aged from 3.5 to 6 years were randomly divided into two groups. Group Ⅰ was ketamine group, group Ⅱ was ketamine plus diazepam group. Ketamine concentrations were determined by reverse-phase high performance liquid chromatography and pharmacokinetic parameters were calculated. Results: The two atrioventricular models were fitted to open two - compartment model. The pharmacokinetic parameters were as follows: group Ⅰ T1 / 2α = (0.15 ± 0.04) h, T1 / 2β = (2.84 ± 0. 44) h, CL = (0.34 ± 0.06) L / kg · h, Vd = (0.66 ± 0.29) L / kg. In group Ⅱ, T1 / 2α = (0.14 ± 0.06) hT1 / 2β = (3.92 ± 0.52) h, CL = (0.26 ± 0.05) L / kg · h, Vd = 0.65 ± 0.07) L / kg. Conclusion: Diazepam prolongs the elimination half-life of ketamine and reduces the clearance rate
