双根清脑颗粒对血管性痴呆模型大鼠神经干细胞增殖迁移的影响

目的:观察双根清脑颗粒对血管性痴呆(VD)模型大鼠神经干细胞(NSCs)增殖、迁移的影响,探讨双根清脑颗粒治疗VD的机理。方法:SPF级SD大鼠42只,随机分为双根清脑颗粒高、中、低剂量组,脑复康组、模型组、假手术组、正常组等7组,每组大鼠6只。采用注入同种血栓栓子方法造成VD模型,中药高、中、低剂量组分别以双根清脑颗粒36,12,4 g.kg-1 ig,脑复康组以0.108 g.kg-1 ig,给药容积均为1.5 mL.kg-1,模型组、假手术组和正常组以等量生理盐水ig。免疫组化法分别标记5-溴脱氧尿苷(BrdU)阳性细胞和微管相关蛋白(DCX)阳性细胞,显微镜下观察BrdU与DCX阳性细胞的表达。结果:双根清脑颗粒中、低剂量组、脑复康组大鼠皮质区与SGZ区BrdU阳性细胞数均明显增加(P<0.05);双根清脑颗粒中、低剂量组SGZ区BrdU阳性细胞比脑复康组增加更为显著(P<0.01)。假手术组以及双根清脑颗粒中、低剂量组大鼠SGZ区DCX阳性细胞数明显增加(P<0.01),高剂量组、脑复康组大鼠SGZ区DCX阳性细胞数亦显著增加(P<0.05);双根清脑颗粒中、低剂量组大鼠SGZ区DCX阳性细胞数比脑复康组明显增加(P<0.01)。结论:双根清脑颗粒可促进VD模型大鼠脑内NSCs的增殖与迁移。 Objective: To observe the effect of Shuanggan Qingnao granule on the proliferation and migration of neural stem cells (NSCs) in rats with vascular dementia (VD) and to explore the mechanism of Shuanggan Qingnao granule in treating VD. Methods: Forty-two Sprague-Dawley (SD) SD rats were randomly divided into seven groups: Shuanggen Qingnao granule high, medium and low dose groups, Naofukang group, model group, sham operation group and normal group. . Using the same method of thrombus embolism caused by VD model, high, medium and low dose groups were treated with Shuanggan Qingnao granules 36,12,4 g.kg-1 ig, Naofu group 0.108 g.kg-1 ig, administration volume was 1.5 mL.kg-1, the model group, sham-operated group and normal group with the same amount of normal saline ig. BrdU-positive cells and microtubule-associated protein (DCX) positive cells were labeled by immunohistochemistry, and the expression of BrdU and DCX positive cells was observed under the microscope. Results: The number of BrdU positive cells in the Shuanggen Qingnao granule group, the low dose group and the Naofukang group were significantly increased (P <0.05); BrdU The positive cells increased more significantly than those of Naofukang (P <0.01). The number of DCX positive cells in the SGZ of sham operation group and Shuanggan Qingnao granule was significantly increased (P <0.01), and the number of DCX positive cells in SGZ of high dose group and Naofukang group was significantly increased P <0.05). The number of DCX-positive cells in SGZ of Shuangen Qingnao Granules was significantly higher than that of Naofukang (P <0.01). Conclusion: Shuanggen Qingnao granule can promote the proliferation and migration of NSCs in the brain of VD model rats.
