目的 研究溶剂汽油对皮肤细胞的损害作用机制。方法 采用皮肤角蛋白细胞和成纤维细胞原代培养技术 ,以及3H 胸腺嘧啶核苷 (3H TdR)、3H 脯氨酸 (3H Pro)、3H 亮氨酸 (3H Leu)和1 4C 亚油酸掺入实验方法 ,观测溶剂汽油对皮肤细胞DNA、蛋白质及皮脂合成能力的影响。结果 接触0 0 1 %的溶剂汽油后 ,角蛋白细胞的3H TdR、成纤维细胞的3H TdR和3H Pro的掺入受到明显抑制 ,抑制率分别为 68 5 %、45 1 %和 40 6 % ;而在接触 0 0 0 1 %溶剂汽油时 ,角蛋白细胞的3H Leu和1 4C 亚油酸的掺入就已表现出显著性降低 ,抑制率分别为 2 0 2 %和 41 2 %。皮肤细胞的掺入抑制率与接触溶剂汽油的剂量有关。结论 溶剂汽油对皮肤细胞具有一定的毒效应 ,可干扰皮肤细胞的正常生理代谢过程 ,影响其DNA、蛋白质及脂质合成能力 ,使皮肤各项生理功能受到影响 ,这可能是溶剂汽油造成皮肤损害的机制之一。与成纤维细胞相比 ,角蛋白细胞更易受到溶剂汽油的影响
Objective To study the mechanism of solvent gasoline on the damage of skin cells. Methods Primary dermal keratinocyte and fibroblast cell culture techniques were used in combination with 3 H thymidine (3H TdR), 3 H Pro, 3 H Leu and 1 C 4 linoleic acid The experimental method was used to observe the effect of solvent gasoline on the DNA, protein and sebum synthesis of skin cells. Results The incorporation of 3H TdR into keratinocytes, 3H TdR and 3H Pro into fibroblasts was significantly inhibited with 0 0 1% solvent gasoline, the inhibitory rates were 68 5%, 45 1% and 40 6%, respectively. However, the incorporation of 3H Leu and 1 4C linoleic acid into keratinocytes showed a significant decrease with the rate of inhibition of 202% and 41 2%, respectively. Skin cell incorporation inhibition rate is related to the dosage of solvent gasoline. Conclusion Solvent gasoline has a toxic effect on skin cells, which can interfere with the normal physiological metabolic process of skin cells, affecting its DNA, protein and lipid synthesis ability, affecting various physiological functions of the skin, which may be caused by solvent gasoline skin damage One of the mechanisms.
