目的 检测脑胶质瘤组织中端粒酶活性及其亚单位的表达 ,研究端粒酶活性与各亚单位表达的相关性。方法 对 6例正常脑组织和 5 1例胶质瘤组织利用端粒重复序列扩增技术 (TRAP)检测了端粒酶的活性 ,利用PT PCR法对端粒酶不同亚单位的表达情况进行研究。结果 端粒酶活性检测 6例正常脑组织均为阴性 ,5 1例胶质瘤组织中 19例阳性。hTR亚单位在 6例正常脑组织中 5例为阳性 ,在 5 1例胶质瘤标本中 48例为阳性。hTRT亚单位在全部正常脑组织中为阴性 ,在 5 1例胶质瘤中 2 1例呈阳性。TPI亚单位在 6例正常脑组织中均呈阳性 ,在 5 1例胶质瘤中 38例呈阳性。端粒酶阳性与hTRT亚单位表达存在明显相关 ,而与hTR和TP1亚单位表达不相关。结论 部分脑胶质瘤中存在端粒酶的活性 ,而hTRT可能是端粒酶激活的限速亚单位
Objective To detect the telomerase activity and subunit expression in glioma tissue and to study the correlation between telomerase activity and subunit expression. Methods The telomerase activity was detected in 6 normal brain tissues and 51 glioma tissues by telomeric repeat amplification protocol (TRAP). The expression of different subunits of telomerase was studied by PT PCR . Results Six cases of normal brain tissue were negative for telomerase activity, and 19 cases were positive for 51 cases of glioma. Five hTR subunits were positive in 6 normal brain tissues, and 48 were positive in 51 glioma specimens. The hTRT subunit was negative in all normal brain tissue and 21 of 51 gliomas were positive. The TPI subunits were all positive in 6 normal brain tissues, and 38 of 51 gliomas were positive. There was a significant correlation between telomerase positive and hTRT subunit expression, but not with hTR and TP1 subunit expression. Conclusions There is telomerase activity in some gliomas, while hTRT may be the telomerase activation rate-limiting subunit
