目的建立高效阴离子色谱-脉冲安培检测法(HPAEC-PAD)测定木糖含量的方法。方法用CarboPacTMPA10分析柱(250mm×4mm),CarboPacTMPA10保护柱(50mm×4mm),ED电化学检测器,Au工作电极,Ti对电极,Ag/AgCl复合参比电极,糖标准四电位波形,淋洗液为50mmol/L NaOH溶液,柱温30℃,流速0.6mL/min。结果木糖在2.00~16.00μg/mL与峰面积呈良好的线性关系(r2=0.9993),最低检测限为0.008μg/mL,平均回收率为100.84%,RSD为1.00%。结论 HPAEC-PAD法操作便捷、无需衍生、分离效果好、灵敏度高,可用于木糖的含量测定。
Objective To establish a method for the determination of xylose content by high performance anion chromatography-pulse amperometric detection (HPAEC-PAD). Methods CarboPacTMPA10 analytical column (250mm × 4mm), CarboPacTMPA10 guard column (50mm × 4mm), ED electrochemical detector, Au working electrode, Ti counter electrode, Ag / AgCl complex reference electrode, The solution was 50mmol / L NaOH solution, the column temperature was 30 ℃ and the flow rate was 0.6mL / min. The results showed that the linear range of xylose was 2.00 ~ 16.00μg / mL and the peak area was linear (r2 = 0.9993). The detection limit was 0.008μg / mL. The average recovery was 100.84% and the RSD was 1.00%. Conclusion The HPAEC-PAD method is convenient and does not need derivatization. It has good separation effect and high sensitivity and can be used for the determination of xylose content.
