目的研究DNA氧化损伤修复相关基因hOGG1和MGMT在正常人支气管上皮细胞(Human bronchial epithe-lial,16HBE)、反式7,8-二羟-9,10-环氧苯并芘(BPDE)转化细胞和裸鼠成瘤细胞中的表达。方法采用原位杂交技术检测hOGG1和MGMT在正常16HBE、反式BPDE转化细胞和裸鼠成瘤细胞中mRNA水平的表达。结果hOGG1、MGMT mRNA在反式BPDE转化细胞中阳性表达明显增强,而在裸鼠成瘤细胞中阳性表达相应减弱。结论DNA损伤修复相关基因hOGG1、MGMT mRNA在转化细胞模型中的表达水平明显增强。
Objective To investigate the effects of hOGG1 and MGMT on DNA oxidative damage repair in human bronchial epitheliial (16HBE), trans 7,8-dihydroxy-9,10-epoxybenzo [a] pyrene (BPDE) And nude mouse tumorigenic cells. Methods The in situ hybridization was used to detect the expression of hOGG1 and MGMT in normal 16HBE, trans BPDE transformed cells and nude mouse tumorigenic cells. Results The positive expression of hOGG1 and MGMT mRNA in trans-BPDE transfected cells was significantly increased, while the positive expression of hOGG1 and MGMT in tumor-bearing cells of nude mice decreased correspondingly. Conclusion The expression levels of hOGG1 and MGMT mRNA in DNA damage repair model were significantly increased in transformed cell model.
